Clonazepam
Indications/Uses
As a single or additional drug in the treatment of Lennox-Gastaut syndrome. Patients w/ absence seizure (petit mal) who have not responded to succinimide.
Dosage/Direction for Use
Adult & Childn >10 yr (>30 kg) Initially 1-2 mg daily. Infant & childn <10 yr (≤30 kg) Initially 0.01-0.03 mg/kg daily. Maintenance Dose: Adult 2-4 mg daily. Chidln 10-16 yr (>30 kg) 1.5-3 mg daily. Infant & childn <10 yr (≤30 kg) 0.05-0.1 mg/kg daily. Max dose: 20 mg daily.
Contraindications
Hypersensitivity. Alcohol or drug dependence, acute narrow angle glaucoma, liver disease. Acute pulmonary disease, resp tract disorders, muscle weakness, myasthenia gravis.
Special Precautions
Chronic resp tract disorders. Avoid alcohol consumption; abrupt w/drawal. May impair the ability to drive vehicle or operate machinery. Liver & kidney diseases. Pregnancy, lactation. Childn.
Adverse Reactions
Fatigue, drowsiness, tiredness, dizziness, lightheadedness, ataxia. Resp depression on IV route. Increased salivation & bronchial secretion. W/drawal symptoms (after long-term use).
Drug Interactions
Increased serum level w/ phenytoin or pyrimidone. Combination w/ valproic acid may cause petit mal seizure. Concomitant use w/ barbiturate, hydantoin, or carbamazepine. Synergistic effect w/ other anticonvulsants, anaesth & hypnotic agents.
